What is AOD-9604?
AOD-9604 was developed as a modified fragment of human growth hormone intended to retain a fat-metabolism signal without reproducing the wider growth-hormone effects. The idea is biologically interesting; the human obesity programme is also a useful warning that a plausible mechanism and a successful weight-loss treatment are not the same thing.
Why people are interested
The potential benefits, unpacked.
Here are the goals and claims that bring readers to this guide. The evidence note shows which part is demonstrated, early research or still a question.
Body-fat reduction
The intended research goal; larger obesity evidence has not established reliable weight reduction.
Body recomposition
A community interest, not an outcome proved by a growth-hormone connection.
Targeting one area of fat
A claim requiring direct evidence; the reviewed trials do not establish spot reduction.
What is it actually doing?
AOD-9604 is based on the C-terminal region of human growth hormone, commonly described as a modified hGH 176–191 fragment. It was designed around laboratory observations suggesting this region could influence fat metabolism without producing the growth-promoting effects associated with intact growth hormone.
That gives AOD-9604 a tidy hypothesis: separate the proposed lipolytic part of growth hormone from the rest of the hormone. What matters clinically, however, is whether that translates into meaningful loss of body weight or body fat in people. The later obesity programme did not establish that outcome convincingly.
Human evidence
What did the studies actually find?
participants reported in the pivotal Phase 2b obesity programme
The larger placebo-controlled programme did not show convincing separation from placebo and obesity development was discontinued.
mean loss in an earlier 12-week 1 mg/day arm reported in secondary literature
Placebo lost about 0.8 kg, but the effect was not dose-dependent and the later larger trial failed.
good evidence for spot reduction
Neither the obesity programme nor the mechanism establishes that injected AOD selectively removes fat from one chosen body area.

OPTIONS Phase 2B obesity study — FDA-reviewed development record
Randomised, double-blind, placebo-controlled · 502 randomised adults with obesity · 24 weeksParticipants received oral AOD-9604 0.25 mg, 0.5 mg or 1 mg once daily, alongside a dietician-supervised diet and exercise programme. The trial was powered to detect a 1.8 kg advantage over placebo at the primary endpoint. It did not find a statistically significant weight-loss advantage for AOD-9604.
This is the study that matters most when somebody says AOD-9604 is a proven fat-loss treatment: the largest dedicated obesity trial was negative, and the developer subsequently terminated the obesity programme.
What it doesn't prove: The FDA review notes that full methods and preliminary efficacy results were not located in the published medical literature, so we should not invent precise arm-by-arm weight-loss figures that are not securely reported.

Randomised dose-ranging obesity study · 300 adults · 12 weeksThe earlier programme produced a small signal. FDA's review reports a mean weekly weight-loss rate of −0.22 kg/week for the 1 mg group versus −0.07 kg/week for placebo, while other reported dose groups did not show a simple dose-response pattern. The FDA characterised the differences as small and of unclear meaningful therapeutic effect, with limited methodological detail available.
This is a good example of why an early positive-looking signal must survive a larger confirmatory study. Here, it did not.
What it doesn't prove: The FDA review says it could not locate a full publication of the study methods and results. The available abstract-level data are incomplete.

What doses and schedules are actually documented?
Human research or licensed product information is separated from clinic and community practice. These are source-attributed reference protocols, not personalised dosing recommendations.
Human research / licensed or product dosing
| Protocol / context | Amount & frequency | Duration | Titration | Evidence |
|---|---|---|---|---|
| OPTIONS Phase 2B | 0.25, 0.5 or 1 mg orally once daily | 24 weeks (primary endpoint 12 weeks) | No titration; parallel fixed-dose arms | Human RCT / company trial record |
| Earlier 12-week study | 1 mg/day oral highlighted in review; earlier development explored higher doses | 12 weeks | Fixed dose | Human RCT summarized in review |
Community / clinic / specialist protocols
| Pattern | Amount & frequency | Cycle | Progression | Status |
|---|---|---|---|---|
| Common SC convention | 300–500 mcg once daily | Often 8–12 weeks | Usually fixed; some start 250–300 then move to 500 | Community; route not validated by obesity trials |
The largest 24-week programme failed to show significant weight loss versus placebo; oral trial dosing should not be used as proof for SC community protocols.
Evidence checked: 7 October 2026
Routes and formulations matter. A topical study does not validate injection; an IV research dose does not establish a fixed SC dose; and evidence for one molecular form must not be silently transferred to another.
Community reports
What are people actually doing?
Daily microgram schedules are commonly discussed
Reddit peptide communities frequently discuss roughly 250–600 micrograms per day, sometimes with 5-days-on/2-days-off variations. These are community protocols, not validated clinical regimens.
View the discussion on Reddit ↗Results are decidedly mixed
Community reports include people describing no measurable benefit after weeks or months as well as people who believe AOD helped alongside diet, training or other drugs. That makes attribution especially difficult.
View the discussion on Reddit ↗The community claims run ahead of the trial evidence
Discussions about fasting, injection timing, local fat targeting and stacking are common, but they should not be presented as established effects simply because a protocol is repeated online.
View the discussion on Reddit ↗Safety, side effects & what we don't know
Published early human work reported generally reassuring short-term tolerability and did not show the IGF-1 or glucose changes expected from full growth hormone in those studies.
That does not establish long-term safety of grey-market injectable AOD-9604. Product identity, purity, sterility and formulation can introduce risks that a controlled pharmaceutical trial does not answer.
The largest practical evidence problem is efficacy: the dedicated obesity development programme did not produce convincing enough weight-loss results to continue development for obesity.
Questions worth asking
Does AOD-9604 burn belly fat specifically?
There is no good human evidence that it can choose abdominal, thigh or any other local fat depot. Spot-reduction claims need direct evidence and the obesity trials do not establish it.
Is AOD-9604 the same thing as HGH?
No. It is a small modified fragment based on part of the growth-hormone sequence, developed specifically to avoid behaving like intact growth hormone.
Why is it still popular if the obesity programme failed?
The mechanism is appealing, peptide communities keep discussing it, and negative or unpublished development programmes are much less visible online than successful modern weight-loss trials. Popularity is not evidence of efficacy.
